Inhibition of D-amino acid oxidase by aromatic acids.

چکیده

Benzoate competes with n-amino acids for n-amino acid oxidase and thus inhibits the activity of the oxidase (l-3). Monosubstituted benzoates, depending upon the substituent, have higher and lower affinities for the oxidase than has benzoate (2). Kojic acid, a Y-pyrone, likewise competitively inhibits the oxidase (4). The present report describes the inhibition of n-amino acid oxidase by certain acids, particularly heterocyclic acids having aromatic properties; e.g., acid derivatives of furan, pyrrole, and pyrones. Like the benzoic acids (l-3), kojic acid (4), and 4-7-carbon, straight chain, fatty acids (5), the substances compete with n-amino acids for the oxidase and are in contrast to the aromatic nitrogen compounds that compete with the flavin component of the oxidase (3). The findings suggest some analogies between the inhibitors and the amino acids that bear upon the interaction of these substances with the oxidase.

Inhibition of D - Amino Acid Oxidase

In an earlier paper from this laboratory (1) there were described the characteristics of competitive inhibition resulting from the interference of certain aromatic compounds with the association of flavin adenine dinucleotide and the separated protein of n-amino acid oxidase. Included among the inhibitors were auramine, quinine, and various other quinolines and anilines. We now present our find...

Role of D-tryptophan oxidase in D-tryptophan utilization by Escherichia coli.

Mutants of Escherichia coli K-12 that require L-tryptophan (trp) are normally unable to utilize D-tryptophan to fulfill their requirement. However, secondary mutations (dadR) that confer this ability can be isolated. In such strains two distinct enzymes are found to be produced at high levels: D-amino acid oxidase (EC 1.4.3.3) and D-tryptophan oxidase. A convenient assay procedure for D-tryptop...

Design and Synthesis of New Benzimidazole and Pyrimidine Derivatives as α-glucosidase Inhibitor

In an endeavor to find a novel series of antihyperglycemic agents, new benzimidazole and pyrimidine derivatives were successfully synthesized efficiently in high yield with high purity, starting from amino acids in the presence of phosphorus oxychloride (POCl3). The synthesized compounds were identified by 1H-NMR, 13C-NMR, FT-IR spectroscopic techniques and elemental analysis. All products were...
